From Microorganism
EC 1.5.3.1

Cat. No.

CED023

Description

Sarcosine oxidase (SAO) is an enzyme that catalyzes the oxidative demethylation of sarcosine to yield glycine, H2O2, 5,10-CH2-tetrahydrofolate in a reaction requiring H4-tetrahydrofolate and oxygen.

sarcosine + H2O + O2 = glycine + formaldehyde + H2O2

CAS No.

9029-22-5

Synonym

L-Pipecolic Acid Oxidase; Sarcosine-Oxygen Oxidoreductase (Demethylating); Oxidoreductases, N-Demethylating

Physical form

Yellowish powder, lyophilized

Specific Activity

≥8U/mg protein

Unit definition

One unit causes the formation of one micromole of hydrogen peroxide per minute at pH 8.0 at 37℃.

Molecular Weight

65 kDa (Gel filtration)

Isoelectric point

5.0

Michaelis constant

2.7×10-3M(Sarcosine)

Optimum pH

8.0

Inhibitors

Ag+, Hg2+, Cu2+ are strongly inhibiting. The even stronger inhibiting effect of thiol group suggests that SDS might play a role in the active site of the enzyme.

Preparation Instructions

The enzyme is reconstituted in 20mM Tris-HCl buffer, pH 8.0 for activity assay.
