From Calf Spleen
EC 3.5.4.4

Cat. No.

CED001

Description

Adenosine deaminase (ADA) is a purine catabolic enzyme ubiquitous in mammalian tissue. In vitro it catalyzes deamination of both adenosine and 2'-deoxyadenosine to inosine and 2'-deoxyinosine respectively.

Adenosine + H2O  →  Inosine + NH3

CAS No.

9026-93-1

Synonym

Adenosine aminohydrolase; deoxyadenosine deaminase.

Physical form

Suspension in 3.2 M (NH4)2SO4, 0.01 M potassium phosphate, pH 6.0;

Specific Activity

150~200U/mg protein

Unit definition

One unit will deaminate one micromole of adenosine to inosine per minute at pH 7.4 at 25℃.

Molecular Weight

32.5~33 kDa (SDS-PAGE)

Isoelectric point

pH 4.85

pH Stability

4.0~10.0

Composition

The enzyme is a glycoprotein containing galactosamine and glucosamine. Its composition shows a predominance of acidic amino acids.

Optimum pH

7.4

Optimum temperature

75

Storage Temperature

2-8℃(Stable≥6 months)

Thermal Stability

<60 (pH7.4, 30min)

Inhibitors

Ag+, Hg2+, Cu2+, sulfhydryl reagents (pCMB and methyl mercuric nitrate) are strongly inhibiting. The even stronger inhibiting effect of TPCK and DFP suggests that histidine or serine or both might play a role in the active site of the enzyme.

Preparation Instructions

The enzyme preparation may be diluted in water or buffer to a protein concentration of 1mg/ml, producing a clear, colorless solution.
